Biomolecules and nanostructures

The Optical Sciences group studies the interaction of light and matter at the nanoscale. We do this by exploring ways to shape light and its environment. It's what we call active and passive control. Our current focus is on the interaction of light with biomolecules and nanostructures. We are part of Twente University's Department of Science and Technology and member of the MESA+ institute.
We participate in the EU-COST actions MP1102: Coherent Raman microscopy (MicroCor) and CM1202: Supramolecular photocatalytic water splitting (PERSPECT-H2O)

 

Near field optical microscopy and spectroscopy




The aperture type Near-field Scanning Optical Microscope (NSOM), realised in 1993, has been improved and applied in the study of several physical, biophysical and chemical structures:

  • Polarised fluorescence of Langmuir-Blodgett mono-layers showing the molecular orientation of the polymer backbone and degree of polymerisation on nm-scale.

  • Detection of fluorescent DNA probes on human metaphase chromosomes with a resolution superior to conventional epi-fluorescence microscopy. A resolution of better than 80 nm has been realised on metaphase chromosomes while a topographic image (50 nm resolution) is obtained simultaneously with the optical image. To our knowledge this is the first useful biological application of NSOM.

  • First detection of Raman spectra with a near-field probe on diamond and polystyrene films by long time integration using a photon counting CCD camera.

  • Fluorescence of single molecules with the near-field scanning optical microscope has been observed and is the main item of current investigations.


  • This FOM-BR project has been concluded with a thesis in 1995 and is continued by a new project focused on "Photodynamics of Single Molecules", financed by FOM-AQ following the 1994 evaluation.
